Dazu muss sich der Spieler wieder durch die Ruinen zahlreicher Testkammern der Aperture Science Labors puzzeln. Um sich einen Weg durch die tödlichen Chambers zu bahnen, verschießt Chell Portale mit der Portal-Gun und setzt viele neue Hilfsmittel wie Beschleuningungs- und Sprung-Gel ein. Außerdem gibt es jetzt Traktorstrahlen, Lichtbrücken und mit Spiegelwürfeln umlenkbare Laserstrahlen in den Testkammern. Die wichtigste Neuerung ist aber der Koop-Modus mit den beiden humorigen Robotern Atlas und P-body.
